Chemotherapy, multidrug resistance (MDR), its modulation and relationship of these events to programmed cell death in human tumor cells

The goal of the submitted project is: A. in vitro analysis of effects and mechanisms of acting selected clinically actual anticancer- and chemopreventive drugs on sensitive and mutidrug resistant (MDR) human tumor leukemia, lyphoblastoid and carcinoma cells with defined mechanisms of multidrug resistance B. with the aid of the cytofluorometric methods, biochemical a immunochemical techniques, to investigate some key processes of the reaction of tumor cells with selected chemotherapeuticals and chemopreventive drugs - proliferation, cell cycle, expression and activity of transported and untrasported plasma membrane proteins of tumor cells (transmembrane and mitochondrial cell programmed death protein molecules), to study signaling pathways after treatments with the selected drugs (single and/or in combinations) in the process of multidrug resistance, its inhibition/modulation, as well as in apoptosis C. evaluation and correlation the results with clinical data determined in related tumors „in vivo“ („short-time“ cell tumor cultures)
