Molecular markers for prediction of individual radiosensitivity in cancer therapy

This study will be performed with cultured human cancer cell lines and blood samples from breast cancer patients with a goal to develop sensitive assay for prediction of individual radiosensitivity of patients undergoing cancer therapy. Analysis of gamma-H2AX DSB repair foci by confocal microscopy has been used as a most sensitive technique for detecting DSB and estimation of repair capacity. Recent data indicate that other molecular markers may provide a better accuracy and reliability Therefore, we will analyze these molecular markers and study their correlation with DSB repair and radiosensitivity using complementary techniques. Analysis of molecular markers for DSB repair in nuclear matrix by flow cytometry and Western blot may be more useful in clinics. These techniques will be applied to study effects of radiotherapeutic doses of ionizing radiation in human cells by comparison of molecular endpoints suggested here with radiosensitivity of cells and the score for individual radiosensitivity of patients.
