Molecular marcers of DNA repair in oncology

While analysis of g-H2AX repair foci with fluorescent microscopy has been used as a most sensitive technique for detecting DSB and estimation of individual radiosensitivity, we and others have recently reported that other molecular markers of DSB repair may provide a better accuracy and reliability. Therefore, relevant phosphorylated and non-phosphorylated forms of DSB repair proteins will be analyzed to the aim of optimizing DSB detection in human cells. Alternative measuring the DNA repair foci by flow cytometry and micro plate reader will be evaluated. DSB repair foci will be analyzed in cultured human cells and lymphocytes from cancer patients undergoing radiotherapy in order to develop new sensitive assay for prediction of individual radiosensitivity of patients. While radiosensitization by hyperthermia has well been established, the mechanisms of this phenomenon remain unclear. Using optimized DSB detection, we will evaluate the role of DSB in hyperthermia
